For patients with newly diagnosed multiple myeloma not undergoing an autologous stem cell transplant, indefinite lenalidomide maintenance resulted in more toxicity with no survival benefit from continuing treatment until disease progression compared with stopping after 2 years. Results from the randomized phase III ENDURANCE clinical trial by the ECOG-ACRIN Cancer Research Group (ECOG-ACRIN) were published today in The New England Journal of Medicine.
As reported in The Lancet by Lu et al, an interim analysis of the Chinese phase III HARMONi-6 trial showed that ivonescimab plus chemotherapy was associated with significantly improved overall survival vs tislelizumab plus chemotherapy in patients with previously untreated advanced squamous non–small cell lung cancer (NSCLC). A prior report from the trial showed that progression-free survival, the primary endpoint, was significantly better with ivonescimab plus chemotherapy.

On July 14, the U.S. Food and Drug Administration (FDA) granted traditional approval to selpercatinib (Retevmo), an oral RET inhibitor, for adult and pediatric patients aged 2 years and older with locally advanced or metastatic solid tumors with a RET gene fusion, as detected by an FDA-approved test, whose disease has progressed on or following prior systemic treatment or who have no satisfactory alternative treatment options. Selpercatinib received accelerated approval for this indication for adult patients in 2022 and for pediatric patients aged 2 years and older in 2024.
